They also show that changes in the pulsations caused by high blood pressure slow the removal of waste, reducing its efficiency.<\/p>\n<p>This might explain the association between high blood pressure and Alzheimer\u2019 disease, the researchers say. Alzheimer\u2019s, the most common cause of dementia among older adults, is characterized by abnormal clumps and tangled bundles of fibers in the brain.<\/p>\n<p>The study, reported in <a href=\"https:\/\/www.nature.com\/articles\/s41467-018-07318-3\"><em>Nature Communications<\/em><\/a>, builds upon groundbreaking discoveries about the brain\u2019s waste removal system by Maiken Nedergaard, co-director of the University\u2019s <a href=\"https:\/\/www.urmc.rochester.edu\/ctn.aspx\">Center for Translational Neuromedicine<\/a>. Nedergaard and her colleagues were the first to describe how cerebrospinal fluid is pumped into brain tissue and flushes away waste. Subsequent research by her team has shown that this glymphatic waste removal system is more active while we sleep and can be damaged by stroke and trauma.<\/p>\n<p>This latest research shows \u201cin much greater depth and much greater precision than before\u201d how the glymphatic system functions in the perivascular spaces that surround arteries in the outer brain membrane, says <a href=\"http:\/\/www.hajim.rochester.edu\/me\/people\/faculty\/kelley_doug\/index.html\">Douglas Kelley, an assistant professor of mechanical engineering<\/a> and an expert in fluid dynamics. (Ä¢¹½´«Ã½ illustration \/ Jeff Tithof)<\/figcaption><\/figure>\n<p>For this study, Humberto Mestre, a PhD student in Nedergaard\u2019s lab, injected minute particles in the cerebrospinal fluid of mice, and then used two-photon microscopy to create videos showing the particles as they moved through the perivascular spaces.<\/p>\n<p>Jeff Tithof, a postdoctoral researcher working with Kelley, then used an automated particle tracking code developed by the Kelley lab that can follow a dot from frame to frame of a video, and after a few frames estimate its velocity and acceleration. From a typical video spanning \u201ctens of minutes,\u201d the code could track more than 20,000 individual particles and derive about a million total measurements to quantify their movements, which show up as \u201ccomet tails\u201d behind each of the tracked dots in the videos.<\/p>\n<p>By analyzing videos from experiments involving multiple mice, \u201cwe were able to gather a really rich data set of what\u2019s actually happening with this flow,\u201d Kelley says.<\/p>\n<p>The researchers found that:<\/p>\n<ul>\n<li>The flow of cerebrospinal fluid is synchronized with the heartbeat, as seen in the animation below. \u201cOne of the main points of this paper is that this flow is mostly \u2013 and maybe purely &#8212; driven by the flexing of the artery wall,\u201d Kelley says.<\/li>\n<li>When high blood pressure was induced in the mice, the flow slowed down and was not as efficient. Artery walls are muscular; they have to flex harder in order to hold the same shape when there\u2019s more pressure inside, and they get stiffer,\u201d Kelley says. \u201cAnd that changes the waveform of the flexing of the artery wall.\u201d<\/li>\n<\/ul>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignright size-full wp-image-360472\" src=\"https:\/\/www.rochester.edu\/newscenter\/wp-content\/uploads\/2019\/01\/cerebrospinal-fluid.gif\" alt=\"animated gif illustrating cerebrospinal fluid flows\" width=\"331\" height=\"345\" \/><\/p>\n<p>\u201cThat&#8217;s highly interesting because early onset high blood pressure is known to be a risk factor for Alzheimer\u2019s in humans,\u201d Kelley says. \u00a0\u201cThere are still a few steps between what we did and chronic high blood pressure. But it may be that high blood pressure\u2014by reducing the pumping of cerebrospinal fluid so that waste isn\u2019t cleared out as well &#8212; is one mechanism that leads to Alzheimer\u2019s.\u201d<\/p>\n<p>The findings should resolve a debate among researchers about whether the flow of cerebrospinal fluid in the glymphatic system is in the same direction as arterial blood flow, as demonstrated by this paper, is opposite, or is random molecular diffusion, Kelley says. \u201cThis is going to quell a lot of those debates.\u201d<\/p>\n<p>The collaboration with Nedergaard also includes Department of Mechanical Engineering faculty members John Thomas (a coauthor of this paper) and Jessica Shang, along with Ali Ert\u00fcrk with the Institute for Stroke and Dementia Research at Ludwig Maximilians University of Munich. Other coauthors of this paper include Ting Du, Wei Song, Weiguo Peng, Amanda Sweeney, and Genaro Olveda of Nedergaard\u2019s lab.<\/p>\n<hr \/>\n<h3><strong>Learn more about Rochester\u2019s role in understanding the brain&#8217;s waste removal system.<\/strong><\/h3>\n<p>Maiken Nedergaard was recently awarded the 2018\u00a0<a href=\"https:\/\/www.lunduniversity.lu.se\/article\/neurology-researcher-wins-prestigious-prize-for-discovery-of-brains-cleaning-system\">Eric K. Fernstrom Foundation Grand Nordic Prize<\/a>\u00a0for her work that led to the discovery of the brain\u2019s unique waste removal system and its role in a number of neurological disorders. Nedergaard maintains labs at the Medical Center and the University of Copenhagen.<\/p>\n<p>In 2012, <a href=\"https:\/\/www.urmc.rochester.edu\/labs\/nedergaard.aspx\">Nedergaard\u2019s lab<\/a> was the first to reveal the brain\u2019s unique process of removing waste, dubbed the glymphatic system, which consists of a plumbing system that piggybacks on the brain\u2019s blood vessels and pumps cerebral spinal fluid (CSF) through the brain\u2019s tissue, flushing away waste.<\/p>\n<p>Nedergaard\u2019s lab has since gone on to show that the glymphatic system:<\/p>\n<ul>\n<li><a href=\"https:\/\/www.urmc.rochester.edu\/news\/story\/3956\/to-sleep-perchance-to-clean.aspx\">works primarily while we sleep<\/a>,<\/li>\n<li>could be a\u00a0<a href=\"https:\/\/www.urmc.rochester.edu\/news\/story\/3870\/brains-garbage-truck-may-hold-key-to-treating-alzheimers-and-other-disorders.aspx\">key player in diseases like Alzheimer\u2019s<\/a>,<\/li>\n<li>is\u00a0<a href=\"https:\/\/www.urmc.rochester.edu\/news\/story\/4213\/blows-to-head-damage-brains-garbage-truck-accelerate-dementia.aspx\">disrupted after traumatic brain injury<\/a>,<\/li>\n<li>may be\u00a0<a href=\"https:\/\/www.urmc.rochester.edu\/news\/story\/5268\/in-wine-theres-health-low-levels-of-alcohol-good-for-the-brain.aspx\">enhanced by moderate alcohol consumption<\/a>, and<\/li>\n<li>could be harnessed as a new way to\u00a0<a
